Long chain PUFAs cannot be synthesized by human body. Therefore, have to be taken from diet only. The present study aims to determine fish oil contents from different parts of fish (head, body and tail) using Bligh & Dyer method and followed by optimization the extraction conditions (temperature, pressure and extraction time) of fish oil from selected part of Tongue sole using Supercritical Fluid CO2 Extraction. The results showed a significant difference (p <0.05) in percentage of fish oil yield between samples. A major amount of lipids were accumulated in body (4.46 ±0.06%) when compared with remaining parts like head (4.29±0.01%) and tail (3.83±0.02%). Therefore, the body part of Tongue sole was a representative part of the fish for optimization using supercritical fluid CO2 extraction (SC-CO2). The use of SC-CO2 offers suitable extraction that promising for food and pharmaceutical industry. However, the proper sample preparations have to be done carefully in order to achieve good results. In the present work, the moisture content of dried ground fish and particle size were 6% and 0.5 mm, respectively. In this moisture level and particle size, the extraction ability of oil was not affected. Process optimization was conducted using central composite design (CCD) for extractions condition using SC-CO2. Independent variables were operating temperature (35 to 75 ºC), pressure (150 to 450 bars) and extraction time (45 to 120 min). The response and variables were fitted well to each other by multiple regressions. The result showed that pressure, extraction time as well as the interaction between pressure and extraction time, temperature and extraction time showed significant effects on the oil yield. The optimal extraction conditions for oil yield (with the experimental range of the variables reached) was at 300 bar, 55 ºC and 82.50 min. Under these conditions, the oil yield was 4.11%. The content of PUFAs in oil extracted was up to 28.47%. Its properties come up to the required standard for edible oil. | ||
