This study aimed to investigate the effect of Tinospora crispa aqueous extract (TC) on elevated blood glucose, anti-oxidant and oxidative stress level in streptozotocin (STZ)-induced diabetic rats. Fifty mg/kgbw of STZ was intraperitoneally induced to Sprague-Dawley rats. The animals were divided into five groups (n=6), as follows: normal control (N), normal TC treated (NT), diabetic control (D), diabetic TC treated (DT), and diabetic treated with standard commercialized Vitamin E (DE). Treatment was delivered for four weeks and blood sample were withdrawn for measurement of those variables. The data among groups were compared using one way ANOVA test, while paired T test was used to analyse the differences between data of pre and post treatment. Diabetic rats showed a significant increase in the blood glucose after STZ induction (16.38±0.54 mmol/L) compared to the normal rats (4.98±0.03 mmol/L; P<0.05). This elevated blood glucose remained steady till the end of study for group D and DE while the administration of TC in DT group reduced the blood glucose level considerably (P<0.05). The result of N group did not differ noticeably with NT group and the body weight normally rose. In diabetic groups, the body weight decreased considerably throughout the study, however body weight of DT group slightly increased after 2 weeks administration of TC. The antioxidant status in DT rats showed that TC prevented a decrease of superoxide dismutase level which was reduced in other diabetic groups. It also increased the glutathione peroxidase enzymes in animals TC treated (NT and DT). Oxidative stress levels (lipid hydroperoxide and isoprostane) went up substantially in group D and no significant changes in other groups. It showed that TC not only has antidiabetic effect but also protected diabetic subject by enhanced anti-oxidant enzymes and prevent the generation of reactive oxygen species. | ||
