Summary: | Diabetes is one of the major diseases that affect most of the world population and in Malaysia 2.6 million people are affected with this disease. The source of our sugar from sugar cane is the contributing factor to the onset of this disease. Hence, one of alternative is from Stevia rebaudiana leaves. The leaves contain steviol glycosides which are compounds that give the sweet taste yet at very low calories. Stevioside and rebaudioside A are two main compounds of this plant that give the sweetest taste approximately 300 times sweeter than sugar cane. In this study, eight accessions of Stevia and tetraploid plants, RAAMBO/01 were planted in similar conditions. The transcript levels of the three genes involved in the production of stevioside and rebaudioside A in all of the accessions and RAAMBO/01 were compared and quantified using real-time quantitative polymerase chain reaction (RT-qPCR). The three UDP-dependent glycosyltransferases (UGT) genes were UGT85C2, UGT74G1 and UGT76G1. The content of stevioside and rebaudioside A in all the varieties were also measured via High Performance Liquid Chromatography (HPLC). For UGT85C2, the highest expression was showed by Taman Pertanian accession, Bertam variety for UGT74G1 and Taman Pertanian accession for UGT76G1. MS 007 had the highest content for both stevioside and rebaudioside A. Based on the correlation study, it was discovered that the transcript level of UGT74G1 has no correlation with stevioside content. The result was similar for UGT76G1 transcript level and rebaudioside A content. Based on the transcript level of genes and stevioside and rebaudioside A content, Bertam, Taman Pertanian and MS 007 were the top three accessions that have the sweetest leaves. These varieties could be considered as sequence of potential commercial value. |
