Summary: | The heavy-chain and light-chain variable region genes of two anti-dengue hybridoma cell line (4G2 and MV8/1/B4-1) were isolated by polymerase chain reaction (PCR) and joined by a flexible peptide linker (Gly4Ser3), as a single-chain variable fragment (scFv) DNA gene. The scFv DNA gene were cloned into a phagemid vector (pCANTAB 5 E) and expressed as a fusion protein with M13 phage in E.coli TG1. Affinity-selection was performed to select and enich scFv phages with strong binding to their respective antigens. |
